ExCLzyme-EN® (Group Companies of Specialty
Enzymes and Biochemicals Co., Chino, California 91710 and
Advanced Biochemicals Ltd. Thane 400601.) belongs to the
class of “Proteolytic enzymes”. The enzymes inhibit
release of pain inducing amines and facilitate drainage of
exudates thereby promoting tissue repair process. They break
down fibrin and inhibit the gathering of platelets around
the healing site and protect tissue from damaging. The
objective was to study anti-inflammatory and analgesic
activity of ExCLzyme-EN®.
Carrageenan induced hind paw edema method of Winter et al,
(1962) in rats was used. ExCLzyme-EN® (100 mg/kg
and 200 mg/kg, p.o.) showed a dose dependant inhibitory effect
(49.73 % and 52.41 % inhibition).
Acetic acid induced writhing method of Taber et al, (1969)
was used to study peripheral analgesic activity in mice. ExCLzyme-EN® (40 mg/kg and 80 mg/kg, p.o.) showed a significant and dose
dependant inhibitory effect (82.73 and 93.01% inhibition).
Carrageenan induced edema is biphasic mediated by release
of histamine and 5-hydroxytryptamine (5-HT) during first
hour, and kinine release up to 2 1/2 h. From 2 1/2 to 6 h
the mediator appears to be prostaglandin, release of which
is closely associated with migration of leukocytes into inflamed
site. ExCLzyme-EN® showed peak inhibitory effect at
3 h indicating inhibition of prostaglandin phase.
It is concluded that ExCLzyme-EN® posses significant anti-inflammatory
and peripheral analgesic activity. |
